摘要

Abstract

[ Objective ] The study aimed to isolate the exocellular β-glucosidase from Aspergillus niger that could hydrolyze the swertiamarin and research its molecular weight, optimal catalytic temperature, optimal pH, stability and catalytic characteristics. [ Method ] The A. niger liquid was fermented for 3 d and then its culture liquid was collected. After the culture liquid was made through the salting out and column separation, a β-glucosidase was gained. [ Result] The β-glucosidase had the molecular weight of 86 kD, the optimal pH of 5.0 -6.0, the optimal reaction temperature of 50 -60 ℃ and showed stable at below 60 ℃. The Ca2+ ,Zn2+ , Fe3+ and Cu2+ could restrain the activity of the βglucosidase and the Mg2+ and Mn2+ could activate its activity. Cellobiose was the optimal catalytic substrate of β-glucosidase. [ Conclusion] This enzyme, similar to the most A. niger from the fungi, showed the weak affinity to the swertiamarin and the strong affinity to p-NPG.
