Abstract

Objective To build an in vitro model of cholesteatoma epithelium proliferation using HaCaT cells, and to investigate the role of p38 mitogen -activated protein kinase (p38MAPK) signal transduction pathway in the development of cholesteatoma. Methods HaCaT cells were incubated with cholesteatoma debris extract (CDE) to produce an in vitro model of cholesteatoma. Expression of p38MAPK in HaCaT cells was examined by immunohistochemistry and RT-PCR. The impact of SB203580, a p38MAPK inhibitor, on HaCaT cell proliferation was also examined by methyl thiazolyl tetrazolium assay(MTT). Results HaCaT cell proliferation showed does-dependent rise 48 hours later. Phospho-p38MAPK positive staining appeared in the nucleus about 30 min after exposure with CDE, but not in non-exposure specimens. Gel image gray-scale analysis (RT-PCR) readings were 0.216 ±0.027 in the control group, 0.828 ±0.019 in the original CDE exposure group, and 0.S22 ± 0.023 in the 1 : 2 attenuated exposure group. The readings in the two exposure groups were higher than the control group (P < 0.05). MTT assay showed effects of SB20358O on HaCaT cell proliferation following exposure to CDE. The mean OD values in the control group, 2.5 junol/L SB203580group, 5u,mol/L SB203580 group, 10 u,mol/L SB203580 group and 20 u,mol/l SB203580 group were 0.535 ± 0.008, 0.527 ± 0.012, 0.504 ± 0.017, 0.476 ± 0.012 and 0.470 ± 0.013 respectively. There was significant difference between the 5 junol/L, lOjunol/L, 20 jimol/1 exposure groups and control group (P<0.05). Conclusion We believe that p38MAPK is activated in the pathologic process of middle ear cholesteatoma. The p38MAPK signal transduction pathway may play an important role in the proliferation of cholesteatoma epithelium.
