摘要

Abstract

Objective To identify effects of the replication-deficient recombinant adenovirus carrying miR-138 on the expression level miR-138 and the target gene hTERT. Methods The human gastric cancer cell line BGC-823 were infected with the replication-deficient recombinant adenovirus carrying miR-138 by the MOI of 200 : 1. The expression levels of miR-138 and hTERT mRNA were inuestigated by real-time PCR. The expression of hTERT protein was detected by western blot. Results Compared to the negative control, after infecting the replication-deficient recombinant adenovirus carrying miR-138 , BGC-823 cells expressed higher level of miR-138 (9. 152 ± 2. 641 vs. 0. 928 ± 0. 199,P<0. 05). No significant difference was observed between the ZERO group (BGC-823 infecting no adenovirus) and negative control (1. 085 ± 0. 478 vs. 0. 928 ± 0. 199,P>0. 05). The expression of hTERT protein was down-regulated in the BGC-823 cells after infected adenovirus carrying miR-138. Conclusion The replication-deficient recombinant adenovirus carrying miR-138 can up-regulate the expression of miR-138 effectively and suppress the expression of hTERT protein.
