Quercetin induces apoptosis of human cervical carcinoma Hela cells

Abstract
Objective: To investigate the cell growth inhibition and apoptosis induced by quercetin on human cervical carcinoma Hela cells and the role of mitochondrium membrane potential in the process of apoptosis. Methods: The cell growth inhibiting ratio of Hela cells was assessed by the acid phosphatase assay. Morphologic changes of the quercetin-treated Hela cells were observed by fluorescence microscopy after acridine orange/ethidium bromide (AO/EB) staining. The cell mito-chondrial membrance potential was detected by flow cytometry. Results: Quercetin inhibited the proliferation of Hela cells significantly, and the growth inhibitory rate presented time and dose-dependent relationship. Typical morphological changes of cell apoptosis were observed after the Hela cells had been exposed to quercetin for 48 hours by the using AO/EB staining method. The cell mitochondrial trans-membrance potentials in Hela cells was declined after being treated with 100 μmol/L quercetin for 24 and 48 hours compared with the control group (P<0. 01). Conclusion: Quercetin can inhibit the proliferation of Hela cells,and induce cell apoptosis by declining trans-membrance potentials of mitochondrium.关键词
