Abstract

Objective:To detect the changes of blood-retinal barrier (BRB) following acute high intraocular pressure (AHIOP) in a rat model. Methods: The rats were randomly divided into normal groups, experimental control groups and experimental groups. The rats of the experimental groups were made into AHIOP model, and then survived for 3 h, 12 h, 1 d, 3 d and 7 d. 3% Evans blue (EB) was injected into the saphenous vein of rats before they were sacrificed. The distribution of EB in the retinal wholemount and retinal slice was observed by using confocal microscopy and the EB content of retina was measured by using spectrophotometer. Results: EB red fluorescence spots were mainly distributed in the ganglion cell layer around capillaries following AHIOP. In 3 h, 12 h and 1 d, it was also found in the inner nuclear layer surrounding the capillaries. The EB red fluorescence spots of the central retina began to increase in 3 h following AHIOP, reached its peak in 12 h, and then gradually decreased. Most of them in the peripheral retina were found in 3 h after AHIOP, and then gradually decreased. Further quantitative analysis showed the same spatial and temporal changes. Conclusion : BRB damage of rats is more evident in its early stage after AHIOP, and restores gradually in its late stage. The serious breakdown of BRB in the peripheral retina precedes the central retina. BRB damage is mainly distributed in the ganglion cell layer.
