白细胞介素-4和白细胞介素-12对纯化培养小鼠视网膜神经节细胞存活的影响
Effect of interleukin-4 and interleukin-12 on the survival of purified mouse retinal ganglion cells
摘要
Abstract
Objective To investigate the effect of interieukin-4 (IL-4) and interleukin-12 (IL-12) on survival of purified retinal ganglion cells ( RGCs) , and to explore the association of autoimmune dysregulation with RGCs injury. Methods Mouse RGCs were isolated and purified using anti-mouse Thy1. 2 monoclonal antibody (two steps of antibody adhesion). The purity of RGCs were verified by immunohistochemistry using double labeled Thy1. 2 monoclonal antibody combined with microtubule-associated protein-2 ( MAP2) polyclonal antibody. Terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining method was used to detect apoptotic cells and the cell number was counted by cell morphology. The present experiment comprised of two stages; Stage 1, to observe the direct effect of IL-4 and IL-12 on RGCs; Stage 2, the influence of IL-4 and IL-12 on RGCs in the presence of NMDA excitotoxicity. Results Except 10 U/ mL IL-4, the survival of RGCs at different IL-4/IL-12 concentrations was significantly lower than that of the normal group (P < 0.0083). In the presence of NMDA, survival of RGCs at various IL-4/IL-12 concentrations (1 U/mL, 10 U/mL and 100 U/mL) was 79.2% , 87.2% and 69.5% ; 77.5% , 76.4% and 73.7% , respectively, all significantly higher than those of NMDA alone treatment group 62.25% ( P < 0.0083 ). Conclusions IL-4 and IL-12 do not show any direct neu-roprotective effect on purified RCCs, but can enhance their action against the NMDA excitotoxicity. Autoimmune dysregula-tion may injure RCCs, and is associated with the pathogenesis of glaucoma.关键词
