p16INK4A蛋白及HPV感染与宫颈上皮内瘤变和宫颈鳞癌的关系研究
Correlations between p16INK4A expression and HPV infection to cervical intraepithelial neoplasia and cervical squamous cell carcinoma

Abstract
Objective:To investigate the relationship between pl6INK4A protein,HPV infection and development of squamous carcinoma of the cervix, by detecting the expression of pl6INK4A protein and HPV infection in tissues of chronic cervicitis cervical intraepithelial neoplasia and squamous carcinoma of the cervix. Methods: p16INK4A protein expression were evaluated in 114 cervix tissues by immunohistoehemical PV-9000. 21 HPVDNA were evaluated by flow-through hybrization and gene chip ( Hybrimax) . Results: (1) In Tissues of chronic cervicitis, CIN Ⅰ ,CIN Ⅱ ,CIN Ⅲ and SCC, the expression of pl6INK4A protein was 0% ,45.83% ,72.73% , 84.00% ,91.30% ,respectively(P<0. 05). With the raising of cervical lesion grade,the positive rate and strength of pl6INK4A increased. (2) In Tissues of chronic cervicitis,CIN Ⅰ ,CIN Ⅱ , CIN Ⅲ and SCC,the expression of HPV was 20% ,37. 50% ,54. 55% ,56.00% ,73. 91% ,respectively (P<0.05). The positive rate of HPV increased. In different cervical tissues,the top three hypotypes of HPV:CIN Ⅰ :HPV16,18,58;CIN Ⅱ ～ Ⅲ :HPV16,33,52;SCC:HPV16,18,52. (3) In Spearman analysis, there was a positive correlation between the expression of pl6 protein and HPV in cervix tissues (r= 0. 268 ,P<0. 05). Conclusion:p16INK4A is related to the grade of cervical lesion. pl6INK4A possibly participated in HPV related formation of cervical cancer. Combined detection has an important significance in screening and prevention of SCC.关键词
