Abstract
[ Objective ] To establish a method for determining contents of chlorogenic acid in different organs from Taraxacum mongolicum by HPLC. [Method] The HPLC was carried out with Shim-pack VP-ODS column (4.6 mm × 150.0 mm,5 μm) using mixture of methanol and 0.4 % phosphoric acid solution in different ratio as mobile phase. Other conditions were: flow rate of 1.0 ml/min, detection wavelength of 326 nm, column temperature of 40 ℃ and sampling amount of 20 μl. [ Result ] When the concentration of chlorogenic acid ranged from 0.791 2 to 79.120 0 μg/ml, the linear relationship between concentration and peak area was good (r =0.999 8). The recovery was 98.00% -101.50%. Leaves、 roots、 flowers and scapes of Taraxacum mongolicum were 1.212, 0.536, 0.448 and 0.417 mg/g in chlorogenic acid, respectively. [ Conclusion] The method established in this work was a rapid and accurate method for determining chlorogenic acid from Taraxacum mongolicum. Leaf of Taraxacum mongolicum was highest in chlorogenic acid, which provided basis for better developing and applying of Taraxacum mongolicum.关键词
