Abstract
OBJECTIVE: To establish a method for the determination of the concentration of doxofylline in human serum. METHODS: RP-HPLC method was adopted. The sample was determined on Diamonsil C18 column with mobile phase consisted of methanol-water (30:70) at a flow rate of 1.0 mL·min-1. The column temperature was 30℃ and the detection wavelength was set at 272 nm. RESULTS: The linear range of doxofylline was 0.101 6~8.120 ng·mL-1(r=0.999 8)with the lowest detection concentration of 0.011 ug·mL-1. The recovery rates of doxofylline at low, middle and high concentration were 84.57% ,93.82% and 92.93% , respectively. The intra-day RSD were 4.65% , 7.34% and 2.91% , and inter-day RSD were 4.65% , 8.60% and 3.74% , respectively. CONCLUSION: The method is proved to be simple, accurate and sensitive, and it is suitable for serum concentration monitoring and pharmacokinetic study of doxofylline.关键词
