Abstract
Objective To investigate the growth status of human periodontal ligament stem cells (hPDLSCs) in simulated microgravity in vitro. Methods HPDLSCs were isolated and cultivated, then characterized by immunohistochemis-try of stromal cell antigen-1 ( STRO-1). After 21 days of induction, the results were evaluated by Alizarin red staining and oil' 0' staining. HPDLSCs were co-incubated with microcarrier beads of Cytodex-3, and were placed in rotary cell culture system. Cells morphology and proliferation potential were examined. Results HPDLSCs were cultivated, and growth characteristics and multipotent differentiation were assessed. The results showed that hPDLSCs can be cultured in simulated microgravity environment. On the 1st day (t =5. 590, P =0. 005), the 3rd day ( t = 12. 238, P =0.000) , the 5th day (t = 19.124, P = 0.000), the 7th day (t=35. 103, P =0.000), simulated microgravity statistically promoted the proliferation potential compared with cells in normal gravity environment. Conclusion The simulated microgravity culture system has the potential to be used for the bioengineering reconstruction of the periodontal tissues.关键词
