摘要
Abstract
Objective To identify the interaction site of Tat with Plkl. Methods Different expression vectors of tat and plkl were constructed. The interaction of tat and plkl was determined by GST-pull down and immunoprecipitation assay. The localization of Tat and Plkl was observed by the confocal laser scanning microscopy. Results Tat protein could interact with the amino-terminal kinase domain of Plkl as shown in GST-pull down and immunoprecipitation assay. The co-lo-caliation of Tat and Plkl in the nuclei of HeLa and 293 cells was observed by the confocal laser scanning microscopy. Conclusion Tat protein can bind to Plkl and activate the kinase activity by up-regulating the level of phosphorylation of Plkl, eventually resulting in the disorder of cell cycle.关键词
