Antibacterial activity in vitro of allicin alone and in combination with imipenem or cefperazone-sulbactam against multidrug-resistant Acinetobacter baumannii

Abstract
Objective To evaluate the activity in vitro of allicin alone and allicin combined with imipenem ( IMP) or cefperazone - sulbactam (SCF) on multidrug - resistant Acinetobacter baumannii. Methods Thirty strains of multidrug - resistant Acinetobacter baumannii clinical isolates were selected randomly in the First Hospital Affiliated to Nanjing Medical University from January 2010 to June 2010. Broth microdilution method was used to measure the minimum inhibitory concentration (MIC) of allicin, IMP, and SCF against the Acinetobacter baumannii. The checkerboard method was used to design the combinations of allicin with the other two different drugs, respectively. Broth microdilution method was used to measure the MIC of different - drug - combinations, and the fractional inhibitory concentration index ( FICI) was calculated according to MIC results. The result of the FICI was interpreted as follows; FICI ≤ 0. 5 for synergy; 0. 5 < FICI ≤ 1 for addition; 1 < FICI≤ 2 for no interactions; FICI > 2 for antagonism. Results The MIC range of allicin against Acinetobacter baumannii was 16 - 64 μg · mL-1, MIC50 was 32 μg · mL-1, MIC90 was 32 μg · mL-1. The FICI of allicin - IMP ranged from 0. 31 to 1. 50, the mean value was 0. 85. The FICI of allicin - SCF range from 0. 31 to 1. 50, the mean value was 0. 86. The FICI ≤ 0. 5 of two joint program was both 20% , 0. 5 < FICI ≤1 was 56. 7% , 1 < FICI ≤2 was 23. 3%. Conclusion The allicin had obvious inhibitory effect in vitro on Acinetobacter baumannii experiment; the results of thetwo joint - program - groups had 20% of synergy, 56. 7% of addition, 23. 3% of no - interaction, 0% of antagonism.关键词
