Abstract

Smashing tissue extraction was used to distill the active triterpenoids of anti-HMGBl from AGS, and the quadratic general rotary unitized design was used to optimize the extraction process. Taking the overall score as the target, the influences of ethyl alcohol concentration, the extraction time and the solvent amount on extraction process were studied. Finally, the optimal extraction technological conditions were determined. The triterpenoids in extraction was determined by UV. The results show that the optimal condition is taking 53% ethyl alcohol as the solvent and distilling for 175 s. This craft is stable and the reproduction quality is good (the relative standard deviation (RSD) is 0.77%), also the operation is simple and takes shorter time. Using acankoreanogenin A as comparison, saturation NaHCO3 as solvent and the most suitable wavelength is 218 nm, the liner range is 0.01-0.07 g/L (R=0.999 6) and the recovery of acankoreanogenin A is 98.2% (RSD is 1.23%).
