An Experimental Study on Differentiation of Adipose Stem Cells into Osteoblasts Induced by Exogenous BMP-7 Combined with VEGF

摘要

Abstract

Objective To study die differentiation of adipose stem cells into osteoblasts induced by low concentration of BMP-7 and VEGF for a short period. Methods The sterile inguinal fat of adult Wistar rat was cut. Adipose stem cells were isolated by collagenase I digestion and cultured in vitro,and divided into 4 groups. DMEM/10% FBS.0.1 junol/L dexamethasone,50 umol/L of vitamin C, 10 mmol/L B-gh/c-erophosphate, 100 U/mL penicillin and 100 U/mL streptavidin hormone were added into each group. On this basis, BMP-7 and VEGF (all 5 ng/mL) were added into the first group;BMP-7 (10 ng/mL) was added into the second group;VEGF (10 ng/mL) was added into the third group;glucose DMEM was added into the blank control group. The medium was replaced every three days. Cell growth and transformation were observed at 1,5,7,10 and 14 d. Osteoblasts induced in the four groups were detected by imrnunohistochemistry,ALP staining,alizarin red staining of calcium nodules and MTT. Results Differentiation ability of adipose stem cells into bone cells was gradually decreased from that of the first group to that of the third group. Differentiation ability of the control group was the lowest. Conclusion Compared to using BMP-7, VEGF or basal medium alone .combination of BMP-7 and VEGF can promote the adipose stem cells differentiating into osteoblasts.
