Association between Toll-like receptor 2 single nucleotide polymorphisms and susceptivity of hepatocellular carcinoma

Abstract
Objective To identify the role of TLR2 single nucleotide polymorphisms and susceptivity of hepatocellular carcinoma. Methods A systematic genetic analysis of sequence variants of TLR2 by evaluating five single nucleotide polymorphisms was performed from 211 hepatocellular carcinoma cases and 232 controls. Genomic DNA was extracted from peripheral blood leukocytes and genotyping was undertaken by using the SNaPshot method. The distribution differences of polymorphisms and related risk between the hepatocellular carcinoma group and the control group were analyzed by the Chi-square test. The relationship of polymorphism genotype with the risk of hepatocellular carcinoma was analyzed by logistic regression. Results Genetic variants of rs3804099、rs3804100 of TLR2 were associated significantly with hepatocellular carcinoma susceptivity (P<0.05). Individuals carrying the heterozygous genotypes of rs3804099 and rs3804100 had decreased risk of aequiring hepatocellular carcinoma (OR adjusted by sex and age 0.493, 95%CI: 0.331-0.736; OR adjusted by sex and age 0.509, 95%CI: 0.342-0.759) (P<0.05). The risk of acquiring hepatocellular carcinoma in individuals with haplotype TT decreased significantly (OR adjusted by sex and age 0.524, 95% CI: 0.394-0.697) (P< 0.01); inversely, the risk in individuals with haplotype CC increased (OR adjusted by sex and age 2.743, 95% CI: 1.915-3.930) (P<0.01). Conclusions The genetic variants of TLR2 may be associated with the risk of occurrence of hepatocellular carcinoma.关键词
