摘要

Abstract

Aim To improve the reliability and sensitivity of popliteal lymph node assay( PLNA )in allergen-ic assessment by analysis of multiple popliteal lymph node( PLN ) immune cells surface molecules. Methods A few female BALB/c mice were divided into four groups and subcutaneously injected with positive drug mercuric chloride ( HgCl2 ), streptozotocin( STZ ), 2, 4,6 - trinitrobenzene sulfonic acid ( TNBS ) and D-penicillamine ( D-pen ) into hind foot respectively. Animals were sacrificed on the 5 th day after injection. Single cell suspensions of injection side of the PLN were prepared. Multiple popliteal lymph node immune cells surface molecules were detected by flow cytometry and then the comprehensive changes were investigated. Results Various significant changes of surface moleules on immune cells were caused by HgCl2 and TNBS,while D-pen and STZ were weaker. Conclusion The changes of the immune cell surface molecules of PLN can reveal the allergenic potential of the test. Because the test causes different types and strength of surface molecular changes , the evaluation of allergenicity should be a comprehensive analysis of changes of cellsurface molecules.
