摘要

Abstract

Aim To investigate effects of angelica A3 active component ( A3 ) on cyclooxygenase-2 ( COX-2 ) activity and expression in lipopolysaccharide ( LPS )-induced mouse RAW264. 7 macrophage cells. Methods Prostaglandin E2( PGE2 ) production and COX-2 enzyme activity were investigated by enzyme-line Im-munosorbnent assay ( ELISA ). RT-PCR and Western blot were used to analyze cyclooxygenase-2 mRNA and the protein expression level. Results A3 20,40, 80mg ? L * was shown to markedly inhibit the LPS-in-duced PGE2 production, COX-2 enzyme activity, COX-2 mRNA and protein expression in RAW264. 7 cells ( P < 0.05 ). Conclusion A3 can directly decrease PGE2 production, which may be related to inhibiting COX-2 gene expression.
