摘要

Abstract

Aim To investigate the preclinical pharma-cokinetics of basic fibroblast growth factor monoclonal antibody ( bFGF-mAb ) in mice. Methods bFGF-mAb was prepared and purified. 1251-bFGF-mAb was labeled by chloramine-T ( Ch-T ), Plasma and tissue were taken at present times. Radioactivity was measured in all samples by trichloroacetic acid ( TCA ) precipitation. Data of plasma concentration were analyzed using 3P97 software. Results Radiochemical purity level of 125I-bFGF-mAb was more than 98% , and trichloroacetic acid ( TCA ) precipitation of 125I-bFGF-mAb was ( 90. 8 ± 10. 2 )%. The main pharmacokinet-ic parameters were as follows: 7j_a( 0. 1 ~ 0. 2 h ), 7j_p ( 1. 05 ~ 1. 84 h ) and 7j_7( 81. 6 ~ 90. 3 h ). Heart, liver and lung were the major organs for deposition of 1251- bFGF-mAb. Conclusion The plasma concentration-time course of 125I-bFGF-mAb is well fitted to three-compartment model, and presents specificity of tissue distribution.
