Study of curative effects and the mechanisms of granulocyte-macrophage colony stimulating factor-genetransfected bone marrow stromal cells on treating cerebral infarction

Abstract
Objective To investigate the curative effects and the mechanisms of granulocyte-macrophage colony stimulating factor (GM-CSF)-gene-transfected bone marrow stromal cells ( BMSCs) on treating cerebral infarction. Methods One hundred and thirty-five SD rats were randomly divided into sham operation group (n = 30), control group (n = 30), BMSCs group (n - 30) and GM-CSF-BMSCs group ( n = 45 ). Middle cerebral artery occlusion ( MCAO) models were made by intraluminal suture method; and 24 h later, the serum-free ot-MEM media, BMSCs and GM-CSF-gene-transfected BMSCs were injected into ischemic boundary zone (IBZ) of rats in control group, BMSCs group and GM-CSF-BMSCs group respectively. At 3 d, 7 d and 21 d after the transplantation, the neurological severity score ( NSS) was detected; and the infarct volume of the model rats and the apoptosis in the IBZ were caculated. The survival and differentiation of transplanted cells in IBZ in GM-CSF-BMSCs group were observed by immunofluroescence technique. Results Compared with the control group, the NSS and percentage of hemispheric lesion volumes ( HLV) of BMSCs group at 21 d after the transplantation were significantly decreased (all P <0. 05) , and the number of apoptosis of BMSCs group at 3 d, 7 d and 21 d after the transplantation were significantly decreased (all P <0. 05); the NSS and HLV of GM-CSF-BMSCs group at 7 d and 21 d after the transplantation were significantly decreased (P < 0. 05 - 0.01), and the number of apoptosis of GM-CSF-BMSCs group at 3 d, 7 d and 21 d after the transplantation were significantly decreased (all P<0.01). Compared with the BMSCs group, the NSS of GM-CSF-BMSCs group at 7 d and 21 d after the transplantation were significantly decreased (all P <0. 05); the HLV of GM-CSF-BMSCs group at 21 d after the transplantation was significantly decreased ( P < 0. 05); the number of apoptosis of GM-CSF-BMSCs group at 3 d, 7 d and 21 d after the transplantation were significantly decreased (all P <0.05). The transplanted cells survived in IBZ and expressed the neuronal marker NeuN in GM-CSF-BMSCs group. Conclusion The curative effects of GM-CSF-gene-transfected BMSCs therapy on cerebral infarction is quite well, which may be related to the anti-apoptotic, nerve replancement and protection.关键词
