Abstract

The aim was to compare and analyze the protein expression profiles of extensively drug-resistant (XDR) and multidrug-resistant(MDR) tuberculosis isolates, and identify differentially expressed proteins related to XDR-TB. Total proteins extracted from clinical isolates of XDR-TB and MDR-TB as well as the standard mycobacterium tuberculosis H37Rv were analyzed with two-dimensional electrophoresis (2-DE). The Progenesis SameSpots program (Nonlinear Dynamics Limited, U-nited Kingdom) was used for analyzing the 2-DE images to identify differentially expressed proteins in XDR-TB as compared to MDR-TB and the standard strain, and some selected protein spots were used for the MALDI-MS peptide mass fingerprinting. The results of this study showed that protein expression profile of XDR-TB was significantly different from those of MDR-TB and the H37Rv standard strain. There were twenty-eight differentially expressed protein spots in XDR-TB. Fifteen proteins differentially expressed in XDR-TB were identified by MALDI-MS peptide mass fingerprinting, Protein expression profile of XDR-TB is different from those of MDR-TB and the H37Rv standard strain, and the differentially expressed proteins could be used as potential target proteins for future investigations to study their functions in XDR-TB.
