NGF withdrawal induced neuronal apoptosis in PC12 cells and changes in HRK/MCL-1 gene expression

Abstract
Objective To investigate the effect of withdrawal of NGF on differentiated neuron-like PC12 cells. Methods Models of differentiated sympathetic neuron-like PC12 cells were created with NGF induction of undifferentiated PC12 cells. After withdrawal of NGF, the changes of morphology and ultrastructure of neuronlike PC12 cells were detected with various methods and RT-PCR were used to verified the expression changes of apoptosis-related genes HRK/MCL-1. Results After NGF withdrawal, neuron-like PC12 cells appeared typical neuronal apoptosis phenotype and the apoptosis rate increased with prolonged NGF withdrawal The expression of proapoptotic gene HRK increased and the expression of antiapoptotic gene MCL-1 decreased significantly within 24h of NGF withdrawal, but without obvious changes after 24 h. Conclusions NGF withdrawal was actually able to induce the differentiated neuron-like PC12 cells showed typical neuronal apoptosis , which related to the expression changes of apoptosis-related genes HRK/MCL-1.关键词
