Expression and significance of glial fibrillary acidic protein and vimentin in sciatic nerve chronic constriction models in rat

摘要

Abstract

Objective: To investigate the morphological changes and expressions of associated proteins after astrocytes are activated in the neuropathic pain. Methods: SD rats were randomly divided into two groups: a chronic constriction injury of the sciatic nerve (CCI) model group and a sham operation group. No. 4-5 lumbar spinal cord was taken to make paraffin slices 1 day before operation and 1, 3, 5, 7 days after operation. The expression of glial fibrillary acidic protein (GFAP) and vimen-tin was labelled by immunofluorescence method, and then double immunofluorescent was stained with double immunolabel-ing techinque. Results: The GFAP positive cells in the (XI groups were increased in the number, larger in size, longer in process in comparing to the sham group. The vimentin positive cells were increased in the number. The mean fluorescence intensities of the GFAP and the vimentin were significantly different compared with others. The mean fluorescence intensity of the vimention began to increase in the sham operation groups after operation with statistical significance compared with the day before surgery. Vimentin and GFAP had a good consistence in operative side of the spinal dorsal horn in the CCI groups. Conclusion: In the neuropathic pain, astrocytes in spinal dorsal horn are activated and show pathological changes of proliferation and hypertrophy.
