Abstract
OBJECTIVE: To establish the method for the concentration determination of sodium valproate in human serum. METHODS: The samples were derivatized with 2-Bromo-4' -nitroacetophenone, and HPLC method was adopted. The chromato-graphic column was Shimpack C18(150 mm×4.6 mm, 5 urn) column with mobile phase consisted of methanol-water (80 :20) at flow rate of 0.9 mL·min-1. The colum temperature was 30 °C and the detection wavelength was set at 265 nm. The cyclohexanecar-boxylic acid was used as internal standard. RESULTS:The linear range of sodium valproate was 10-150 mg·L-1(r=0.999 7,w= 6). The minimum detectable concentration was 1 mg·L-.1 The average recovery of relative and extraction was 99.45% -103.24% and 98.12%~100.37%. The RSD of intra-day and inter-day were less than 8%(n=5). CONCLUSION: The method is rapid, simple and accurate, which can be used for serum concentration monitoring of sodium valproate.关键词
