摘要

Abstract

Objective:To investigate the effects of carbon dioxide (CO2) pneumoperitoneum environment with different pressures on invasion ability of cultured human hepatoma carcinoma cells line SMMC-7721 in vitro. Methods; With an in vitro artificial pneumoperitoneum model, human hepatoma carcinoma cells line SMMC-7721 were exposed to CO2-insufflation of three pressure groups(5. 10 and 15 mmHg) and the control group. After exposion, transwell chamber was used to determine the invasion capacity of hepatoma carcinoma cells in vitro. The different time point after exposion, adhesive kit and immunocyto-chemical stain was respectly used to detected the adhesive ability and the expressions of MMP-9 and VEGF of human hepatoma carcinoma cells. Results: The invasion, adhesive ability and The MMP-9 expressions of hepatoma carcinoma cells line SMMC-7721 in 15 mmHg group showed significantly enhanced than in control group(P<0. 05). The VEGF expressions in 15 mmHg and lOmmHg groups showed significantly raised than in control group (P<0. 05). The adhesive ability of cells and the protein expressions of MMP-9 and VEGF rised gradually and reached a peak at 36 h after exposition, then decreased. Conclusions: High pressure CO2 pneumoperitoneum envioronment can significantly promote the invasion ability of hepatoma carcinoma cells line SMMC-7721. The mechanism may due to obviously promotion of the expression of MMP-9 and VEGF.
