脊神经结扎模型大鼠双足痛阈变化与脊髓促炎因子表达的相关性
Relationship between bilateral paw mechanical pain and the proinflammatory expression in spinal cord in a rat model of spinal nerve ligation
摘要
Abstract
Objective; To analyze the emRNAs expression of IL-ip, IL6 in spinal dorsal hom and tie ir effects on mechanical pain sensitivily following spinal neive ligation in rats. M ethods: Healthiy adultmale SD rats were randomly divided into the spinal neive ligatfan group( Tx) , shan operatbn group( S), control group The50% mechanical paw withdrawal 1hresho]d(50% MWT) of left or right feetwas deternined by up-downmefliod; RT-qFCR measured ihemRNA expression levels of IL-1β and IL-6 in left or right spinal dorsal hom at different tine point after surgery. Results; The50% MWT of ipsilateral feet decreased significantly on3 id day after 15 and 16 spinal neive ligation(P <0.001) and reached tieminmun on21 stday (P < 0.001). The mechanical pain sensitivily of the contralateral feet enhanced significantly only on 10 ti day and 14 th day after nerve ligatfan(P <0.01 ,P <0.05). ThemRNA expression of IL-1β and IL-6 upregulated in ipsilateral dorsal hom on7 1h day after nerve ligation when compared to group S or control group ( P < 0.05); onl4 th day, the level of IL-6 mRNA in Ipsilateral side was still higher than group S or control group(P <0.05). The mRNAs of IL-β, IL-6 in contralateral spinal dorsal hom showed no significant increase on 10 th, 14 th day afterneive ligation Conclusion; Unilateral 15 and 16 nerve ligaticn contributed to mechanical hyperalgesia devebpnent of bilateral feet, however, the mechanical pain sensitivity of ipsilateral feet after neive ligation occurred earlier and maintained longer. IL-1β IL-6 mRNA in bilateral spinal dorsal hom showed different characteristic egression in the tine course, they played an mportant role in neuropatiic pain关键词
