藤梨根乙酸乙酯提取物对肺癌A549细胞凋亡的诱导作用

摘要
Abstract
Objective To observe the induced effect of Radix Actinidiae extracts on apoptosis of A549 cell. Methods A549 cells were cultured in vitro, and were given different concentrations of (40 μg/ml, 80 μg/ml, 160 μg/ml) Radix Actinidiae extracts(treatment group) or 0.04% DMSO(control group). DNA agarose gel electrophoresis( AGE) was used to detect DNA ladder band of A549 cell after treatment. Cell apoptosis was measured by TUN EL after treatment. Immuno-histo chemistry and RT-PCR were used to detect the expressions of Survivin protein and mRNA. Results DNA ladder strap atlas were found in treatment group by DNA agarose gel electrophoresis. Apoptosis rate of AS49 cell in each treatment group obviously increased by dose-and time-dependent manner. The expresses of Survivin protein and mRNA in treatment groups were lower than those in control group( P <0.05 ). Conclusions Radix Actinidiae extracts can induce apoptosis of AS49 cell in vitro, its mechanism may be related with regulating the expression of Survivin gene.关键词
