摘要

Abstract

Objective To investigate the expressions of P27 and PCNA on renal tubular epithelial cells in renal ischaemia and reperfusion injury,and evaluate the effect and possible mechanism of HMG-COA reductase inhibiter fluvastatin on expressions of P27 and PCNA in the cells. Methods 30 rats were randomly divided into 3 groups: sham-operated group(n = 10); IRI group (n = 10); fluvastatin treatment group (n = 10). The in vivo study was conducted by establishing rat model of acute ischemic renal failure through ischaemia/reperfusion and sacrificing the rats 24 hours after both renal hila were clamped with small vascular clamps for 60 minutes and released. At 24 hours after reflow, renal morphology was guantified. Also, the expression and localization of P27 and PCNA were examined in kidney sections by immunohistochemistry. Results Our date shown that the extend of renal morphology changes in Fluvastatin treatment group were remarkably less than that in IRI group , Cell Proliferation Index's (PI) in IRI group was the highest among three groups, and significantly higher than that in Fluvastatin treatment group. The immunohistochemistry analysis shown that the expression of P27 was significantly lower than that in sham group , and elevated by Fluvastatin treatment (P<0.01). The level of P27 had negative correlation with PCNA (r = -0.609 ,P< 0.01). Paller's grades had positive correlation with PI (r = 0.872,P < 0.01 )and negative correlation with P27 (r = - 0.713 ,P < 0.01). Conclusion The level of P27 expression decreased and PCNA expression increased at 24 hours after reflow in the rat model of acute ischemic renal failure. Fluvastatin playing a role in renal protection may enhance P27 expression, make cell cycle arrest in Gl phase and help for DNA repairing.
