Abstract
BACKGROUND: Human umbilical cord derived mesenchymal stem cells (HUC-MSCs) have been confirmed to have thecharacteristics of immunologic tolerance in vitro, but there are no relevant allogenic research in vivo. The results and mechanismare lack of consistency.OBJECTIVE: To cultivate HUC-MSCs and research the immunological characteristics of implanted HUC-MSCs in vivo and in vitro.METHODS: HUC-MSCs were obtained by using tissue adhesion method. Mixed-leukocyte reaction (MLR) was performed toevaluate immunologic tolerance in vitro, flow cytometry applied to detect the changes of CD4 and CD8 expression as well as ratioof CD4 to CD8, and ELISA used to detect interleukin 2 (IL-2), IL-10, ?-interferon (IFN-?) levels.RESULTS AND CONCLUSION: MLR results showed a similar trend in the concentration changes of cytokines: the level of IL-10slightly increased after MLR reaction (P < 0.05), while the level of IFN-? Decreased (P < 0.05). The ratio of CD4+/CD6+ in T cellsubsets was decreased to a slight extent after cell transplantation, indicating that HUC-MSCs possessed the characteristics ofallogeneic immunological tolerance and immunoloregulation effect.关键词
