Abstract
Objective To establish a HPLC method for the simultaneous determination of amoxicillin and clavulanate in human plasma. Methods The chromatographic separation was carried out on Phenomenex Luna Cm column(250 mm x 4. 6 mm, 5 |xm) with the mobile phase consisting of acetonitrile NatbPCK buffer(pH =3. 0) (95 :5),the flow rate was 1. 2 mL/min. The detecting wavelength was at 230 nm. Results Clavulanate and amoxicillin were eluted for about 5 min and 9 min. Cavulanate and amoxicillin showed the good linear correlation in the range of 0. 13 - 8. 60 |xg/mL (r=0.999 4) and 0. 21 - 13. 20 g/mL( r =0. 999 8) respectively. The intra-day RSD and inter -day RSD were both less than 15. 0% (n = 5). Conclusion The method is simple, accurate and suitable for the determination of plasma drug concentration of amoxicillin and clavulanate.关键词
