apelin-13 promotes proliferation of vascular smooth muscle cells by downregulating caveolae expression

Abstract
BACKGROUND: Previous results have shown that caveolae is likely to participate in a pel in-13 promotion of vascular smoothmuscle cell proliferation.OBJECTIVE: APJ e a G-protein coupled receptor, and rts ligand is apelin peptide. Previously, we reported PI3K and ERK1Ssignal pathway mediated proliferation of vas cular smooth muscle cells (VSMCs) induced by apelin-13. ThEstudy e to observecaveolae invoked in rat VSMCs proliferation induced by apelin-13 and to determine whether caveolin-1 proteh binds to signalmolecules PI3K, ERK1/2to mediate VSMC proliferation induced by apelin-13.METHODS: VSMCs were prepared from male Sprague-Dawley ratthoracic aorta by the primary-expl ant method. The effect of &cyclodextrin on cell proliferation induced by apelin-13 was measured by MTT assay. VSMCs were incubated with apel'n-13 andB-cyclo dextrin (B-CD), caveolir-1 expression was detected by western blot assay. Formation of high molecular weight poryp rote incomponent including caveolin-1 and PI3K /ERK1/2 was detected by immunoprecipitation.RESULT SAND CONCLUSION: 6-CD(5 mmol/L, 25 hours) significantly enhanced VSMCs proliferation induced by apelin-13(P < 0.05). Treating VSMCs with apelin-13 (0, 1, 2, 4, 8 u mol/L) downregulated apelin-13-induced expression of caveoln-1 andthe effect was distinct at 1μ mol/L<p < 0.05). Pretreatment of the celb with 5 mmoVL B-CD could strength the do wnregulation ofapelin-13-induced caveolin-1 expression (p < 0.05). Apelin-13 may induce dissociation of PI3K(ERK1/2) wrth caveolin-1 inVSMCs compared with the control group. Thesefindings suggest that caveolae t> invoked in apelin-13-induced VSMCsproliferation.关键词
