Abstract
BACKGROUND: Temperature would influence the proliferation and apoptos is of spermatogonial cells cultured in vitro. OBJECTFVE: To investigate the effects of temperature on cell cycle and pro I iteration-related gene c-krtand cyclinD3 of type AMETHODS: The Eclated spermatogenic eels bytaio-step enzymatic digestion were co-curtured withsertoli eels in an incubator at 32 ℃ and 37 ℃ to observe the morphological changes and growth chara cte ristics every day. On the 8h day ofin vitro culture, typeRESULTS AND CONCLUSION: On the 1S| day of in vitm culture.suspending spermatogenic eels which were round or oval and the adherent sertoli eels that we re fusiform or irregular shape were observed in 32 ℃ group and 37℃ group. Starting from the 4th day. The number and growth state of spermatogenic cele were gradualty decreased in the 37℃ group. At the 8h day.the nunfcer of spermatogenic cells in the 37℃ group was significantly lower than that in the 32℃ group (P <0.05). Flow cytometry resuHs showed that at the Sr day of in vitro culture, the proportion of type A spermatogonial eels at S phase in the 32℃ group was significant^ higher than that in the 37℃ group (P<0.05);the relative mRNA and protein expression levee of c-krt. cyclinD3 in type A spermatogonial celE were significantly higherinthe32 "C group than in the 37℃ group (P < 0.05). Results indicate that spermatogenic cells cultured in vito would inhibit the proliferation of type A spermatogonial cells at body cavrty-lketemperature (37℃).关键词
