摘要

Abstract

[Objective] The objective of this study was to explore the conditions of FACE detecting glucomannooligosaccharides. [Method] Konjac glucomanno-oligosaccharides was used as the substrate, the optimal parameters were determined by investigating the effects of 3 kinds of fluorescer, the concentration of resolving gel, the conditions of derivatization reaction and sample impurities on FACE. [Result] The optimal conditions of FACE used in detecting glucomanno-oligosaccharides were as follows: fluorescer ANTS, concentration of resolving gel 25%, content of ANTS 2 uL, derivation reaction temperature 37°C and derivation reaction time 16 h. [Conclusion] These findings show that FACE has a high resolution to glucomannooligosaccharides, and 14 kinds of components were detected by FACE, which provides an effective method for qualitative and quantitative analysis of glucomanno-oligosaccharides.
