强直性脊柱炎患者HLA-B*27基因亚型及序列研究
Study on HLA-B* 27 gene subtype and sequence in Ankylosing Spondylitis patients in Shanghai

Abstract
Objective To study the distribution of the 43 alleles of HLA-B* 27 subtype in Ankylosing spondylitis( AS) patients, and to provide theoretical and practical basis for AS clinical diagnostics at local area. Methods Blood sample from 70 AS patients were analyzed by low resolution PCR-SSP,and 17 cases from 27 samples showing heterozygosity at B locus were randomly chosen for PCR-SBT analysis. And PCR-SBT was also performed to test 3 cases from which exact results can not be obtained because of the allelic homogeneity. Results Fifty patients showed positive HLA-B*2704 (including 31 homozygotes, 19 heterozygotes) ;21 patients showed positive HLA-B* 2705 (including 10 homozygotes.11 heterozygotes), and 1 patient simultaneously expressed heterozygosity of HLA-B*2704/HLA-B* 2705. The total positive rate of HLA-B* 2704 was71.42% .taking 70.41% of the 70 cases;the total positive rate of HLA-B*2705 was 30.01% .taking 29.59% of the 70 cases. Genotype frequency of B*2704 was 0.578 6;B*2705 was 0. 285 7 by the method of direct calculation. Conclusion HLA-B*27 subtypes in AS patients in Shanghai are mainly HLA-B*2704 and HLA-B*2705,among which HLA-B*2704 predominates.关键词
