Abstract

OBJECTIVE: To study the effect of the alkaloids of Pinellia ternata on human hepatocarcinoma cell strain Bel-7402 proliferation. METHODS: Human hepatocarcinoma cell strain Bel-7402 co-cultured with alkaloids of P. Ternata (400, 200, 100 μg· mL-1). MTT method was used to detect the effect of alkaloids of P. Ternate on human hepatocarcinoma cell strain Bel-7402 proliferation. TUNEL technique was used to detect the effect of alkaloids of P. Ternata on human hepatocarcinoma cell strain Bel-7402 apop-tosis, and immunohistochemistry SABC staining method was used to test Bax and Bcl-2 protein expression. RESULTS: The high-dose, medium-dose and low-dose of alkaloids of P. Ternata (400, 200, 100 ng · mL-1) could inhibit the proliferation of Bel-7402 cell after 24 hours treatment, the inhibitory effect enhanced as the concentration increased, the highest inhibitory rate reached 36.98% in dose-response relationship. TUNEL showed that three different doses of alkaloids of P. Ternata could induce Bel-7402 cell apoptosis in dose-response relationship. Three different doses of alkaloids of P. Ternata could increase Bax expression but decrease Bcl-2 expression. CONCLUSION: Alkaloids of P. Ternata can inhibit human hepatocarcinoma cell strain Bel-7402 proliferation and introduce its apoptosis, and its mechanism is associated with up-regulation of Bax and down-regulation of Bcl-2.
