Abstract
OBJECTIVE: To establish the method for the content determination of salidroside in Rhodiola cremlata and Rhodiola fastigiata. METHODS: RP-HPLC method was adopted. The determination was performed on OPH RP-ODS C18(250 mm × 4.6mm, 5 urn) column with mobile phase consisted of 50 mmol·L-1 phosphate buffer solution (pH value adjusted to 7.0 with sodium hydroxide)-methanol (90:10) at the flow rate of 1.0 mL·min-1. The column temperature was room temperature. The detection wavelength was set at 253 nm and injection volume was 10 μL. RESULTS: The linear range of salidroside was 62.5~1 000.0 μg· mL-1 with average recovery of 97.59% (RSD=1.71% , n=6). Linear relationship existed between the content of salidroside in R. Crenulata and its levels. There was littler or no salidroside in R. Fastigiata, so it can't be used as substitutes. CONCLUSION: The method is accurate, sensitive and precise, and it can used for the content determination of salidroside in Rhodiola Rosea.关键词
