Abstract

Objective To investigate the protective effect of extracts of gingko biloba (EGB) on rat neuron during hypoxi-a/reoxygenation (H/R). Methods The cortical nerve cells were cultured in vitro for 10d and randomly divided into 6 groups: the control group, H/R group, EGB761 low-dose group, EGB761 high-dose group, EGB1212 low-dose group and EGB1212 high- dose group. In EGB- treated groups EGB761(0.01 and 0.lmg/L) or EGB1212(0.01 and O.lmg/L) were added in culture media 24h before H/R, respectively. The morphology of neurons was observed with microscopy, neuronal survival rate was examined with MTT assay and LDH was measured with LDH release assay in the culture media collected after hypoxia for 2.5h and reoxy-genation for 24h. Results The neuronal cell was swelling after 2.5h hypoxia and 24h reoxygenation insult. MTT assay suggested the neuronal survival rate deceased to 50% compared to normal condition. LDH level in the H/R group was increased as compared with the control group. In all EGB- treated groups, the survival rate of the neurons was increased and the LDH levels were decreased as compared with the H/R group. The protective effects ofEGB1212 were more prominent than those of EGB761 at the same concentration. Conclusion H/R can induce the damage of neurons in rats and EGB may protect neurons against the insults with a dose dependent manner.
