Abstract
Based on asynchronous-injection alternating merging zone technology, a flow-injection spetrophotometry for fast determination of real protein content in dairy products was established. Sample and coomassie brilliant blue G250(CBBG) solution were loaded into loops simultaneously and automatically, injected into two carriers, respectively, then merged and reacted in manner of sample enwrapping reagent, and the formed blue complex was determined at 595 nm. Optimized conditions were as follows: concentrations of CBBG, NaCl and HCl were 150 mg/L, 0. 1 mol/L and 1. 0% (V/V) , respectively; volumes of the sample and reagent were 150 μL and 30 μL, respectively; length of a reaction coil was 200 cm; total flow rate was 2. 65 mL/min. The linear range of the method was 0. 5 -15. 0 mg/L, relative standard deviation was less than 1. 87% (n=11), its detection limit was 0. 05 mg/L, and analytical speed was 60 samples per hour. Merits of the system were less error, high automation, good reproducibility, fast analysis speed, and less cost. Moreover, the determination results of this method were not interfered by some pseudo protein such as melamine and urea. The method has been applied to the determination of real protein in dairy products.关键词
