Determination of the Content of Shikimic Acid in Leaf of pine needle in Liuzhou City of Guangxi Province

摘要

Abstract

[Objective]The determination of shikmic acid in pine needle and organic solvent extraction and separation method were established. [ Method ] The shikimic acid in leaf of pine needle was extracted and separated and its content was determined with the method of high performance liquid chromatography(HPLC) with the NH2 being taken as separation column;acetonitrile-2%H3P04(90:10) .mobile phase;the temperature of column,25 ℃ ;the flowing velocity,1 ml/min;and the testing wavelength,213 nm. [ ResultsJThe sample of shikimic acid extracted was with less impurity. The linear range of the shikimic acid was in 0.29 -2.88 μg(R2 =0.999 9) ,the average collection was 98.30% ,RSD was 0. 74% (n = 5). [ Conclusion ] The extraction and separation method was suitable for the determination of shikimic acid with high performance and environmental protection and low cost. The average content of shikimic acid in leaf of pine needle was 1.14%.
