Effect of immunomodulating peptide on T lymphocyte transformation and macrophage secretion in BALB/c mice

Abstract
To study the effect of immunomodulating peptide ( PGPIPN ) deriving from β-cacein on spleen T lymphocyte transformation ( LT ) and murine bone marrow macrophage secretion ( BMMs ) in BALB/c mice. Methods The transformation effect of T lymphocyte with different concentrations of PGPIPN was detected by MTT method, and cytokines ( TNF-α and IL-6 ) released from murine BMMs by ELISA method. Results In vitro, LTs were markedly improved by ConA at 5 μg/ml and PGPIPN at different concentrations( P <0. 01 ). However, different concentrations of PGPIPN didn' t enhance spleen LT alone. The results also showed that PGPIPN with li-popolysaccharide ( LPS ) could stimulate BMMs to secrete TNF-a and IL-6, but PGPIPN alone was incapable of stimulating BMMs to release cytokines. Conclusion PGPIPN can promote LTs both in vivo and in vitro, and stimulate BMMs to release cytokines with LPS together in vitro. It suggests that PGPIPN can increase the immune effect of T lymphocytes and macrophages.关键词
