摘要

Abstract

[Objective]To screen the optimum conditions for separation and purification of total saponins from RADIX ASTRAGALI. [Method] The content of total saponins in RADIX ASTRAGALI was determined by ultraviolet spectrophotometry using dynamic adsorption - desorption method aimed at studying the optimum conditions for separating and purifying total saponins from RADIX ASTRAGALI with macroporous resin. [Result] The optimum conditions for separation and purification of total saponins with AB-8 were as follows; extracting concentration of 0.1 g/ml, sampling volume of 3 BV, flow rate of 1.0 ml/min, washing with 4 BV of distilled water first and then desorption with 4 BV of 50% ethanol. Under the above condition, the yield of total saponins reached as high as 52. 1%. [Conclusion]This method was simple and feasible, which could meet industrial requirements.
