Establishment of SYBR Green Ⅰ real-time PCR for rapid diagnosis of porcine circovirus type 2

Abstract
A pair of specific primers target to open reading frame ORF1 gene of pocine circovirus type 2 was designed and a SYBR Green I fluorescent based real-time PCR was developed for the quantization of porcine circovirus type 2. The detection limit of realtime PCR was 82.2 plasmid copies per μL. The melting curve analysis using SYBR Green I dye showed one specific peak with a melting temperature is (85.98 ±0.13) ℃, and no primer-dimers peak presented. No amplification was detected from unrelated DNA samples by this method, such as porcine circovirus type 1, porcine parvovirus virus. Also no amplification was detected from unrelated RNA samples by this method, such as swine influenza virus, porcine reproductive and respiratory syndrome, pseudorabies virus, classical swine fever virus. Fine reproducibility was obtained for detecting plasmid DNA with intra-assay of 0.29% -1.35% " and inter-assay of 0.96% - 2.42%. The real-time PCR method developed in this study will be useful for rapid laboratory diagnosis and epidemiology investigation for porcine circovirus type 2.关键词
