摘要

Abstract

To study the effect of cells cycle arrest and mechanism in human liver cancer HepG-2 which induced by evodiamine. The cytotoxic effect of evodiamine was determined by MTT assay. The influence of evodiamine on human liver cancer' s cell cycle distribution was examined by flow cytometry. Used immunofluorescence and observed by confocal laser microscopy to investigate the changes of tubulin. Treated HepG-2 cell line with different doses of evodiamine ,for 24 hours, the IC50 was 19.62 μmol/L. Evodiamine could induce G2/M phase arrest. Under confocal laser microscopy, the polymeric form of tubulin was changed significantly, blocked the spindle poles formed. Evodiamine could induce cell cycle arrest in human liver cells line HepG-2, the mechanism had effect on polymeric form of tubulin and play the role of anticancer activity.
