Abstract
The effects of experimental conditions, such as injected solution concentration, concentration and volume of elution solution, for column chromatography, and sample solution s pH value, extracting time and number of repetitions on the purification of chlorogenic acid in extracts of waste and inferior tobacco were investigated. The results showed that: 1) the proper conditions of column (glass column 16 mmx200 mm, bed volume 19.8 mL) chromatography by polyamide resin were: concentration of crude chlorogenic acid solution 8.5 mg/mL, elution solution 198.0 mL 25% ethanol; 2) the appropriate conditions of extraction with ethyl acetate were: pH value of sample solution 2.0, extracting twice, extraction time 5 minutes. Under the mentioned conditions, the content of chlorogenic acid in the obtained product reached 82.1% with the yield of 57.6%.关键词
