Abstract

Objective To demonstrate infrastructure of the undersurface of the tectorial membrane using scanning electron microscopy (SEM) to advance understanding of mechanismsin cochlear sound reception. Methods Tectorial membranes from 12 cochleae in 6 guinea pigs were studied under a HITACHI S-4800 scanning electron microscope. Results (1) Imprints from thestereocilia of outer hair cells were clearly visible on the undersurface of the tectorial membrane, seemingly corresponding to tips of the tallest row of stereocilia. The imprints were "W" shaped in the basal turn, but gradually turned into "V" or irregular shapes towards the apical turn. The radiant fibers on the undersurface were interrupted by the imprints which appeared as a string of beads on the undersurface. (2) A deep linear groove was seen at locations corresponding to inner hair cells, which appeared shallower but broader than outer hair cell imprints. The groove widened from the basal to apical turn. (3) The fibers on the undersurface of the tectorial membrane were radially arranged between theouter edge of the tectorial membrane and inner hair cell stereocilia imprints and interrupted by outer hair cell stereooilia imprints. These were fine fibers, interlaced and connected with each other. Fibers between inner and outer hair cell imprints (I.e., areas over pillar cells) were especially thin and uniform looking filaments, whereas those fibers between inner hair cell imprints and the spiral limbus (above the inner sulcus) appeared even finer and longer. Conclusion The imprints on the undersurface of tectorial membrane clearly indicate contact between the membrane and hair cell stereocilia. The size and morphological differences between fibers at different locations may indicate a role of the tectorial membrane in mechanical tuning.
