摘要

Abstract

Objective:HPLC fingerprints of Radix Rehmanniae Preparata steamed in different producing areas were studied contrastively to establish the safety and efficacy quality control methods. Methods; RP - HPLC was used and Acteoside was used as the standard. The chromatographic conditions were as follow;column using Polaris C18 (5 μm,4.6 x250 mm) ,the detection wave length was at 334 nm, the flow - rate was at 1.0 mL/min constantly, column temperature was at 25 ℃ , and the test solutions were accurately injected 20 mL respectively into the column. Mobile phase using solvent A and B in linear gradient elution(0 -40 min,A 10% -20% ;40 -60 min,A 20% -30% ) .solvent A and B were acetonitrile and 0. 1% acetic acid solution respectively. Results; HPLC fingerprints of Radix rehmanniae Preparata steamed in different producing areas were greatly similarity,the. Similarity was up to 0. 9. The analysis was carried out by reproducibility of the test( n = 5 ) according to the relative retention times and the relative peak areas of peaks. The RSD of retention times range from 0. 10% to 0. 41% ,and the others range from 0.14% to 1. 70% . Conclusion:The method was specific and the major characteristic fingerprint peaks were obvious, it suitable for quality identification of radix rehmanniae preparata.
