红花多糖对人肝癌SMMC-7721细胞增殖的抑制作用
The Inhibitory Effect of Safflower Polysaccharide on the Cell Proliferation of SMMC -7721 of Human Liver Cancer

Abstract
Objective; To observe the inhibitory effect of Safflower polysaccharide(SPS) on the cell proliferation of human liver cancer cell line SMMC -7721. Methods; SMMC -7721 cells were treated with different concentrations of SPS. The effect of SPS on the rates of cell proliferation was detected with MTT assay. Morphology changes were observed with inverted microscope. The effect of SPS on apoptosis was detected by FCM, SMMC -7721 cells were double - stained by Annexin - V/PI, and by fluorescence microscope, SMMC -7721 cells were stained by acridine orange(AO). Results: MTT methods indicated SPS had a significantly inhibitory effect on SMMC - 7721 cells in a dose - and time - dependent manner(P <0. 05 ,P <0. 01 ). While cells in SPS group showed shrink, bad refraction, and more dead cells were observed by light microscopy. AO fluorescent staining results showed that part of cell cytoplasm condensed, the volume of narrow, nuclear shrinkage, chromatin increased, more brightly fluorescent, condensed to form dense green fluorescent after 24h. The crescent - shaped shrinkage can be seen in some cells nucleus after 48h and 72h. Apoptotic rate increased with the increase of the concentration of SPS by FCM. Conclusion: SPS can obviously inhibit the growth of SMMC -7721 cells cultured in vitro in a dose effect relationship and time effect relationship. SPS can induce apoptosis of human hepa-toma cell line SMMC -7721 in a dose -dependent manner.关键词
