Abstract
[ Objective ] In order to study the coId toIerance function of Atc0rl5a from Arabidopsis. Method The pIant expression vector of pCAMBIAl30l - CORl5a was constructed and then it was transferred into tobacco genome through agrobacterium tumefaciens. Antibiotic seIection and PCR detection was used for detecting transgenic tobacco. It was to expIore the effect of Iow temperature stress on soIubIe sugar and proIine content of transgenic and wiId - type tobacco by determination of anthrone method and the acid ninhydrin and observe their coId toIerance at the same time. ResultIt was found that soIubIe sugar content and proIine content of transgenic tobacco were higher than those of wiId - type tobacco, and reached peak vaIue at day 5. After4C and -lC stress the transgenic tobacco pIants can grow normaIIy whiIe aII wiId -type tobacco pIants died. [ conclusion ] The determination of the physioIogicaI and biochemicaI data show that the changing trend of soIubIe sugar content and proIine content were positiveIy correIated to coId toIerance of tobacco and the resuIts suggested that expression of Atc0rl5a gene significantIy increased the coId toIerance of transgenic tobacco.关键词
