Abstract

AIM: To study the effects of soybean isoflavones on neuronal apoptosis and mRNA expression of calpain in transient focal cerebral ischemia and reperfusion ( FCIR ) rats. METHODS: Thirty - six adult healthy female SD rats were ovariectomized and randomly divided into soybean isoflavone - treated group and control group. The rats in soybean isoflavone - treated group were subdivided into low and high doses of soybean isoflavone groups. After 1 month of treatment, a FCIR model was established by intraluminal middle cerebral artery occlusion ( MCAO ) with a nylon monofila-ment suture. In situ hybridization was performed to examine the mRNA expression of calpain. Apoptosis was observed with terminal deoxynucleotidyl transferase - mediated dUTP nick - end labeling ( TUNEL ). The activity of calpain was also examined. RESULTS: The mRNA expression of calpain and TUNEL positive cells were observed in the cortex and striatum of ischemic hemisphere. Calpain mRNA was present at lower levels in soybean isoflavone - treated groups than that in control group ( P <0. 05 ). The number of apoptotic cells decreased in soybean isoflavone - treated groups compared with control group ( P < 0. 05 ). No significant difference of the calpain mRNA and the number of apoptotic cells between high -dose and low - dose soybean isoflavone - treated groups was observed ( P <0. 05 ). The activity of calpain was also reduced in soybean isoflavone -treated groups. CONCLUSION: Soybean isoflavones attenuate ischemic neuronal apoptosis by reducing the activity and inhibiting the mRNA expression of calpain.
