地塞米松和1,25-(OH)2D3对EC1细胞增殖和细胞周期的影响
Effects of dexamethasone and 1,25- (OH) 2 D3 on cellular proliferationand cycles of EC1 cells
摘要
Abstract
Aim:To explore the impact of DEX, 1,25-(OH)2D3 and DEX in combination with 1,25-(OH)2D3 on the cellular proliferation and cycles of EC1 cells in different periods of time. Methods :The EC1 cells were treated by 10-7 mol/ L DEX, 10-7 mol/L 1,25-( OH)2D3 and the combinations in same concentration for 48,72,and 96 h. The EC1 cells were treated by absolute ethyl alcohol in same level as the solvent control group. The morphological change of the cells was observed by optical inverted microscopy, the state of cellular proliferation was tested by MTT method, and the distribution of cell cycle was measured by flow cytometry. R6SUlts;The proliferation or cycle of EC1 cell were inhibited significantly in 1, 25-( OH )2D3 group or DEX group (inhibition rate of cell proliferation: F1.25-(OH)2D3 = 51.185,P<0.001; rate of cell staying in G0/G1 phase:FDEX =8. 170,P =0.009) , and for DEX factor, 1,25-(OH)2D3 factor ,time factor,there is interaction between any two factors ( ( inhibition rate of cell proliferation-. F1.25(OH)2D3×DEX=11. 017, P = 0. 003; F1.25-(OH)2D3×tiem 3.668,P = 0.041;FDEX×time= 7.887,P =0.002. Rate of cell staying in G0/G1 phase:F1.25-(OH)2D3×DEX=12.381,P= 0.002;F1.25-(OH)2D3×time =6583,/>=0.005;FDEX×time =5.096, P =0.014) ,but there is not interaction among time and the combination group( inhibition rate of cell proliferation:F1.25-(OH)2D3×dex×time =0. 512,P=0. 606;rate of cell staying in G0/ G1 phase:F1.25-(OH)2D3×DEX×time=0.410,P =0.668). Conclusion:l ,25-( OH)2D3, DEX and their combination could in-hibit the cell proliferation and induce cell cycle to arrest in C0/C1 phase.关键词
