Abstract

Aim; To study the influence of angiopoietin-like protein 3 ( ANGPTL3 ) antisense oligodeoxynucleotide (ASODN) on the ability of invasion and adhesion of EC9706 cells. Methods: The ANGPTL3 ASODN was synthesized based on its gene sequence recorded in the GenBank. EC9706 cells were transfected with mixture liquid of liposome and ASODN, and then to look into the influence of ANGPTL3 ASODN on the adhesion rate, the number of the cells penetrated through the membrane, and their invasion inhibiting rate to the EC9706 cells. Results: After 48 hours, distribution of EC9706 turned more sparse compared with that of the control group. The bulk of the cells transfected by ASODN became small. The crimples were rotundity and the figure refraction of the nucleus and cytoplasm boosted up but became granulate. Part of the cells were suspending in the culture liquid but did not grow against the wall. The adhesion rate of the ASODN group decreased a lot compared with the control and SCODN ones( P<0.05). The number of the cells penetrated through the membrane reduced significantly(P<0. 05 ) but the invasion inhibiting rate increased significantly ( t = 124. 470, P< 0. 001). Conclusion; The ANGPTL3 ASODN could depress the ability of invasion and adhesion of EC9706 cells and consequently affect the malignant biological behavior.
